Der Verteidiger begann seine Profikarriere 2000 bei Independiente Medellín in Kolumbien. 2003 wechselte er für ein Jahr zu den Boca Juniors nach Buenos Aires, um von Argentinien aus nach Europa zu wechseln.

Von 2004 bis 2012 spielte Perea bei Atlético Madrid. Er absolvierte über 300 Spiele für die Rojiblancos und feierte seine größten Erfolge mit ihnen.[1]

Zur Saison 2012/13 wechselte Perea ablösefrei zum CD Cruz Azul.

Perea spielte 76 Mal im kolumbianischen Nationalteam.

Von August 2018 bis Juni 2019 war Perea Trainer des kolumbianischen Vereins Leones FC aus Itagüí.[2][3]

Luis Amaranto Perea Mosquera (Turbo, 30 de enero de 1979) es un exfutbolista y entrenador colombiano nacionalizado español. Actualmente sin club. Dirigió al Cadete C de las divisiones inferiores de Atlético de Madrid y también al Itagüí Leones de Colombia.[1][2] Jugó como defensa central, su retiro se produjo en 2015. Fue el jugador extranjero que más partidos disputó con la camiseta de Atlético de Madrid hasta que fue superado por Diego Godín. A lo largo de los ocho años que pasó en el club rojiblanco jugó 314 partidos, llegó a ser el segundo capitán del equipo y conquistaría dos Europa Leagues en 2010 y 2012 y una Supercopa de Europa en 2010. Junto a Pablo Ibáñez, formó una pareja de centrales recordada en su época.
